Wham Wham’s Redemption is a unique action-adventure platformer featuring a new cast of characters (including yours truly) and dynamic music from indie composer & musician Bonte Avond. In an imaginative, snow-soaked setting, the game’s protagonists, Bonte and a bunch of otherworldly creatures, journey through a wintery fantasyland while fighting to keep their world from ending. With moody, indie soundscapes from Bonte playing in the background, watch out for strange cactus-like plants with a mean attitude, and cryptic characters that will test your logic.
Besides the main game and the bonus post-credits story, there is a music site with information about Bonte, as well as a free download of the game’s soundtrack and a special bonus album.

Bonte is a Dutch electronica and orchestral composer based in Amsterdam. He started making music at age 11 when he picked up a guitar. After watching the movie Breaking Away, he decided he wanted to create music like the game. He played around with his guitar and got quite good. But he wasn’t satisfied.

For all the pain and suffering that flowed from New France, the French who stayed on after the official expulsion of 1629 became the most loyal soldiers of the French empire. The Acadian region of Northern Nova Scotia was one of the last to be settled by Europeans, and the inhabitants became some of the most loyal subjects of Louis XIV. The population of the region quickly became divided into two groups, the French loyalists known as ‘Acadians,’ who wanted independence from the king, and their Protestant ‘Innanis,’ who wanted to remain loyal to the king. This unstable situation allowed for an extraordinary mix of cultures and social structures to develop in the region, and while the physical landscape is similar to the Acadian landscape, the game is entirely fictitious, with no historical characters. You play as one of these fictional characters, making decisions on behalf of his or her clan or family. The identity of your character and clan is determined completely at random. If there is a variant, it’s random. Every time you play it you are a different person with a different set of skills and attributes.

Description:
Charley’s War is a reenactment historical game set during the French and Indian War (Seven Years War).
In it, players take on the role of the French and Indian Native American allies. By gaining victory over their Colonial enemies, they are able to gain control over more land, which in turn allows for the expansion of their civilization. The campaign map encompasses more than 20 square miles of historical terrain. This includes the areas of Fort La Tour, Fort Duquesne, Fort Duquesne (“The Rocks”), and Point Levi. Each player has their own individual village and it is up to them to expand it, or succumb to the English and the rival Native American tribes.

Nether is a game set in a block of apocalyptic metropolis with a population of a million people. It has two playable characters: The Warrior and the Mage, each with their own progression, weapons, abilities and equipment available throughout the game. Nether:

The discovery of massive parallel gRNA libraries was a major breakthrough in CRISPR/Cas9 editing. The widespread use of these libraries enabled a rapid pace of introducing mutations into mammalian genomes. Nonetheless, they are not perfect. As shown in [@bib1], one of the major issues for *in vitro* cell-based systems was that library bias could influence the resulting mutagenic spectrum. Although the accuracy of using *in vitro* gRNA libraries for genome editing could be promising in some cases, the robustness remains to be established.

In a recent paper published in *eLife*, [@bib2] present data suggesting a bottleneck in the CRISPR/Cas9 genome editing process. They report that the use of *in vitro*-selected gRNAs did not lead to the expected increases in the incidence of mutations in targeted regions of the mammalian genome. Instead, they show that the selected gRNAs generate a mutagenic spectrum biased to repeat sequences, such as transposons. This creates concerns about the robustness and reproducibility of the use of *in vitro*-selected libraries for genome editing.

In the study of [@bib2], very few gRNAs were tested *in vivo*. Therefore, the library source is not a concern as long as every single gRNA was selected from a population. On the other hand, the gRNAs tested *in vitro* were introduced in an extrachromosomal plasmid, which could lead to false-positive results.

A major issue in the study of [@bib2] is that, at least to our knowledge, they used 5′-phosphate-dependent crRNAs. These crRNAs have a heterogenous
