Dietary brominated lipids elicit differential responses in the immune system of a monogonont rotifer.
Chronic exposure to brominated flame retardants (BFRs) alters immune system function and the ability of animals to cope with pathogen challenge. Moreover, our previous work shows that BFRs have the potential to alter the microbial communities of animals that can directly affect resistance to pathogens and immunomodulatory responses. In this study, we exposed juvenile southern damselflies (Calopteryx virgo) to environmentally relevant concentrations of brominated lipids (2.5ng/L) for 12days, followed by a challenge with a yeast pathogen (Saccharomyces cerevisiae). We first compared the acute effects of diet on a number of feeding and foraging behaviours, following which we investigated effects on immune system function using a series of innate immune assays (Toll, Prophenoloxidase, Encapsulation and Phagocytosis) and an antibody dependent cellular cytotoxicity assay. We found that BFR exposure did not impact the majority of behaviours examined but significantly decreased the success rate of Encapsulation and Phagocytosis assays, and decreased encapsulation success when combined with yeast exposure. Both immunostimulatory and immunosuppressive effects of diet were observed, with no effect on the innate immune response to yeast when compared to a control diet. In the antibody dependent cellular cytotoxicity assay, BFR exposure significantly decreased the resistance of flies against the yeast pathogen challenge. There was no difference in the response to the yeast challenge when adult damselflies were previously exposed to the diet at different stages of development. However, there was a decrease in immunity in damselflies exposed as larvae when compared to newly emerged damselflies. The results of this study suggest
